Saline buffer

Phosphate-Buffered Saline (PBS) Calculator

Scale a common 10X PBS formulation for bench preparation and dilute to working concentration as needed.

Scientific use note

  • What this tool does: scales common fixed buffer recipes, calculates stock dilution volumes, or calculates mass from molarity, volume, and molecular weight.
  • What this tool does not do: replace institutional protocols, validate sterility, provide clinical or manufacturing-grade validation, calculate arbitrary pH recipes in v1, or replace SDS/MSDS review.
  • Research-use reference only. Verify all calculations against your institutional protocol before use. Check SDS/MSDS documentation for each reagent and follow standard laboratory PPE, handling, and waste-disposal procedures.
  • This page presents static recipe information in the initial HTML and adds local interactive scaling in the browser.
  • Adjust pH slowly using acid or base while measuring with a calibrated pH meter. Follow your lab protocol.

Component Type Amount Notes
NaCl solid 4.00 g Use your lab's standard reagent grade.
KCl solid 100 mg Use your lab's standard reagent grade.
Na2HPO4 solid 720 mg Common anhydrous reference value; verify hydrate form if substituting.
KH2PO4 solid 120 mg Use your lab's standard reagent grade.

Bench protocol

  1. 1.Add about 80% of the target final volume of distilled or deionized water to a clean vessel. Prepare 1X PBS for a final batch of 500 mL.
  2. 2.Add the weighed salts and stir until fully dissolved.
  3. 3.Adjust pH slowly using HCl or NaOH while measuring with a calibrated pH meter, if your protocol requires it.
  4. 4.Bring to final volume with water and mix thoroughly.
  5. 5.Sterilize or store according to your application and lab protocol.

FAQ

Can I use this PBS calculator for 1X or 10X PBS?

Yes. The base recipe is stored as 10X, and the calculator scales component amounts for the target concentration you choose.

Does this tool calculate pH adjustment volumes?

No. BufferPrep does not estimate exact acid or base volumes in v1. Adjust pH slowly while monitoring with a calibrated meter.
