Hybridization buffer

20X Saline-Sodium Citrate (SSC) Calculator

Scale a common 20X SSC stock recipe for nucleic acid hybridization workflows.

Scientific use note

  • What this tool does: scales common fixed buffer recipes, calculates stock dilution volumes, or calculates mass from molarity, volume, and molecular weight.
  • What this tool does not do: replace institutional protocols, validate sterility, provide clinical or manufacturing-grade validation, calculate arbitrary pH recipes in v1, or replace SDS/MSDS review.
  • Research-use reference only. Verify all calculations against your institutional protocol before use. Check SDS/MSDS documentation for each reagent and follow standard laboratory PPE, handling, and waste-disposal procedures.
  • This page presents static recipe information in the initial HTML and adds local interactive scaling in the browser.
  • Adjust pH slowly using acid or base while measuring with a calibrated pH meter. Follow your lab protocol.

NaCl solid 4.38 g Use your lab's standard reagent grade.
Sodium citrate solid 2.21 g Hydration state matters.

Bench protocol

  1. 1.Add water to about 80% of the final batch volume. Prepare 1X 20X SSC for a final batch of 500 mL.
  2. 2.Dissolve sodium chloride and trisodium citrate dihydrate fully.
  3. 3.Adjust pH only if required by your protocol and only while measuring with a calibrated meter.
  4. 4.Bring to final volume with water and mix thoroughly.
  5. 5.Label the stock concentration clearly before storage.

Recipe notes

  • Common formulation; verify against your institutional protocol before use.

FAQ

Why is hydration state mentioned for SSC?

Sodium citrate is often supplied in different hydrate forms, which changes the mass required. Verify the exact reagent form before weighing.

Can I use this for 1X SSC?

Yes. Choose the target concentration and the calculator scales the common 20X stock accordingly.
